Aspergillus tamarii expresses an extracellular alkaline protease that we show to be effective in removing hair from cattle hide. Large quantities
of the enzyme will be required for the optimization of the enzymatic dehairing process so the growth conditions for maximum
protease expression by A. tamarii were optimized for both solid-state culture on wheat bran and for broth culture. Optimal protease expression occurred, for
both cultural media, at initial pH 9; the culture was incubated at 30 °C for 96 h using a 5% inoculum. The crude enzyme was
isolated, purified and characterized using MALDI TOF TOF. The alkaline protease was homologous to the alkaline protease expressed
by Aspergillus viridinutans.

Nothapodytes nimmoniana is a medicinally important tree species that occur in the Western Ghats, a megadiversity hotspot in southern India. Inner stem bark of the tree contains an important anti-cancer alkaloid, camptothecin for which the natural population of the tree is heavily harvested. In this paper, we report the isolation and characterization of eight polymorphic microsatellite loci using enrichment hybridization protocol. Analysis of 36 individuals representing two populations revealed three to 12 alleles per locus. Observed heterozygosity ranged from 0.21 to 0.94 for the two populations. None of the loci tested showed linkage disequilibrium. These markers are invaluable for evaluating the genetic structure and assessing the genetic impacts of harvesting of N. nimmoniana in the Western Ghats to formulate strategies for conservation of the species. 相似文献

The species of the genus Populus, collectively known as poplars, are widely distributed over the northern hemisphere and well known for their ecological, economical, and evolutionary importance. The extensive interspecific hybridization and high morphological diversity in this group pose difficulties in identifying taxonomic units for comparative evolutionary studies and systematics. To understand the evolutionary relationships among poplars and to provide a framework for biosystematic classification, we reconstructed a phylogeny of the genus Populus based on nucleotide sequences of three noncoding regions of the chloroplast DNA (intron of trnL and intergenic regions of trnT-trnL and trnL-trnF) and ITS1 and ITS2 of the nuclear rDNA. The resulting phylogenetic trees showed polyphyletic relationships among species in the sections Tacamahaca and Aigeiros. Based on chloroplast DNA sequence data, P. nigra had a close affinity to species of section Populus, whereas nuclear DNA sequence data suggested a close relationship between P. nigra and species of the section Aigeiros, suggesting a possible hybrid origin for P. nigra. Similarly, the chloroplast DNA sequences of P. tristis and P. szechuanica were similar to that of the species of section Aigeiros, while the nuclear sequences revealed a close affinity to species of the section Tacamahaca, suggesting a hybrid origin for these two Asiatic balsam poplars. The incongruence between phylogenetic trees based on nuclear- and chloroplast-DNA sequence data suggests a reticulate evolution in the genus Populus. 相似文献

We have identified, isolated, and characterized microsatellite/simple sequence repeat (SSR) loci in trembling aspen (Populus tremuloides) by screening partial genomic libraries. We have also examined the compatibility and use of the P. tremuloides SSR primers to resolve microsatellites in other Populus species. Fourteen microsatellites were identified from 1600 clones screened. The TC/AG microsatellites were the most abundant.
A total of 29 alleles were detected in 36 P. tremuloides individuals at the four SSR loci (two each of di- and tri-nucleotide repeats) characterized. The number of alleles at the
SSR loci ranged from 5 to 11, with an average of 7.25 alleles per locus, and the observed heterozygosity ranged from 0.19
to 0.82, with a mean of 0.46 per locus. Although the highest polymorphism was observed for a dinucleotide SSR locus, the trinucleotide
SSR loci showed substantial polymorphism. There were 34 unique multilocus genotypes among the 36 P. tremuloides individuals examined, and 89% of the individuals had unique multilocus genotypes. Two pairs of SSR primers were successful
in PCR, amplifying genomic DNA and resolving microsatellites of comparable size from Populus deltoides, P. nigra, P.×canadensis, and P. maximowiczii. The microsatellite DNA markers developed could be used for clonal fingerprinting, certification of controlled crosses, genome
mapping, marker-assisted early selection, genetic diversity assessments, and conservation and sustainable management of poplar
genetic resources.

Lipid peroxides are considered to be the initiation factor for atherosclerosis. Present study depicts that L-carnitine treatment (300 mg/kg body weight/day) for 7 and 14 days caused significant reduction in the tissue lipid peroxidations. Salt grass is an important pioneer plant in early stages of succession. The sharp-pointed rhizomes with numerous epidermal silica cells, and the aerenchymatous network of the rhizome, leaf sheath, and roots facilitate development of the plant in heavy clays, shales, and inundated soils. In salt marshes of southern Utah, salt grass contributes to a hummock-building process that favors localized removal of salts by capillary action and evaporation. This process provides a narrow strip of soil that is favorable for the rooting of extended rhizomes. In laboratory experiments, maximum growth for Distichlis spicata, a perennial salt marsh grass, was obtained at 15,000 ppm soluble salts in nutrient solution cultures. Comparable concentrations of salts occurred in soils of the habitat from which plants were taken. Nearly equal concentrations of sodium and potassium were found in the plant tissue where the growth of the plants was optimal; such a ratio was maintained in the plants during most of the growing season. In the field the greatest amount of growth of salt grass takes place when temperatures are cool and soil moisture is quite high during the early spring. During mid-summer as air temperatures rise, crude protein in the plant decreases. During periods of high salt and water stress, morphological and anatomical adaptations of the stomata, salt glands, and trichomes of salt grass are important for survival. Stomata on exposed ridges of vascular bundles, where desiccation is greatest, usually are covered by four epidermal cells. In contrast, stomata found in the grooves between vascular bundles tend to be uncovered. The salt gland is composed of a large basal cell and a cap cell and actively excretes (in a diurnal rhythm) excess sodium, potassium, and chloride ions. A mechanism for salt excretion from this gland is postulated. The silica-containing trichomes on the leaves may play a role in cooling the leaf under conditions of high solar radiation and also serve to protect the plant against attack by herbivores. 相似文献

The efficacy of 16, 16 dimethyl PGE2 p-benzaldehyde semicarbazone ester for cervical dilatation prior to evacuation of the uterus in 180 first trimester nulliparae has been studied. The drug was injected into the muscle of the cervix 3 hours before vacuum aspiration. In 143 patients (80%) the cervix had dilated adequately to enable evacuation of the uterus without mechanical dilatation. In the remaining 37 patients (20%) the cervix had dilated to 6 or 7 mm and additional mechanical dilatation could be performed easily in most of these patients. Side effects consisted of vomiting (11%), diarrhoea (7%), transient pyrexia and shivering (7%). There were no complications in any of the patients and no perforation of the uterus or damage to the cervix resulted during evacuation. 相似文献

Myosin-6 is an actin-based motor protein that moves its cargo towards the minus-end of actin filaments. Mutations in the gene encoding the myosin-6 heavy chain and changes in the cellular abundance of the protein have been linked to hypertrophic cardiomyopathy, neurodegenerative diseases, and cancer. Here, we present a detailed kinetic characterization of the human myosin-6 motor domain, describe the effect of 2,4,6-triiodophenol on the interaction of myosin-6 with F-actin and nucleotides, and show how addition of the drug reduces the number of myosin-6-dependent vesicle fusion events at the plasma membrane during constitutive secretion. 相似文献
